Urologist who led the CCGA study validating the Galleri methylation-based multi-cancer detection test.
Eric Klein led the Circulating Cell-free Genome Atlas (CCGA) study that developed and clinically validated the Galleri cell-free DNA methylation test for detecting more than 50 cancer types and predicting tissue of origin, the basis for the PATHFINDER and NHS-Galleri trials. As a urologist he led the SELECT prevention trial and the development of the Oncotype DX prostate score.
A multi-cancer blood test can be run in ordinary clinics, and most positive results can be resolved with imaging. But six in ten positives are false alarms that take months to resolve, and the test misses most cancers. Whether it reduces late-stage cancer or deaths is unknown; that requires randomised trials.
Whether a cancer is found depends mostly on how much tumour DNA it sheds into the blood, which is why small early cancers are often missed by every approach.
Methylation patterns can flag many cancers and point to the organ, but the cancers most worth catching early were the ones least often found.
