Alligator Bioscience is a Swedish biotech that makes antibodies to switch on the immune system inside tumours; its CD40 agonist mitazalimab, given with chemotherapy, shrank tumours in 42 per cent of people with untreated metastatic pancreatic cancer in a single-arm trial, which cannot show whether it improves survival.
Alligator Bioscience, based in Lund, develops tumour-directed immunotherapies from its FIND antibody library and RUBY bispecific format. Mitazalimab (ADC-1013), a CD40 agonist that primes dendritic cells and T cells, was tested with modified FOLFIRINOX in the single-arm phase 1b/2 OPTIMIZE-1 trial in untreated metastatic pancreatic cancer, which met its primary endpoint with a confirmed response rate of 42.1 per cent and reported median overall survival of 14.9 months; with no control group it cannot show whether mitazalimab improves survival, and the authors plan a randomised confirmatory trial. ATOR-4066 is a Neo-X-Prime bispecific against CD40 and CEACAM5.
Mitazalimab is a CD40 agonist antibody from Alligator Bioscience AB, in registered phase 2 trials for pancreatic ductal adenocarcinoma.
