Compass makes antibodies that hit two targets at once to reshape the blood supply and immune environment of tumours. Its lead drug, tovecimig, blocks two blood-vessel growth signals. In September 2026 the FDA advised the company to run a trial showing that it helps people with bile duct cancer live longer before applying for approval, and the company is still preparing to apply.
Compass Therapeutics is a Boston clinical-stage, oncology-focused company developing antibody-based therapeutics. Lead candidate tovecimig is a DLL4 x VEGF-A bispecific antibody; data from the COMPANION-002 phase 2/3 study in biliary tract cancer were selected for an oral presentation at ESMO 2026. In that study tovecimig plus paclitaxel improved objective response rate (18.0% against 5.3% with paclitaxel alone) and median progression-free survival (4.7 against 2.6 months, hazard ratio 0.44), but the overall survival analysis did not reach statistical significance, which the company attributes to crossover from the control arm. On 22 September 2026 Compass announced that the FDA had recommended a trial demonstrating a survival benefit before a BLA for previously treated advanced biliary tract cancer; the company said it does not believe a new trial is warranted and continues to prepare a BLA submission. CTX-8371, a PD-1 x PD-L1 bispecific, is in phase 1 cohort expansions in triple-negative breast cancer, Hodgkin lymphoma and non-small cell lung cancer, and CTX-10726, a PD-1 x VEGF-A bispecific, has dosed its first phase 1 patients with initial data expected in the fourth quarter of 2026. A phase 2 study of CTX-471, a CD137 agonist antibody, in NCAM (CD56)-expressing tumours is planned. Compass had $180 million in cash and marketable securities at 30 June 2026, expected to fund operations into 2028. Nasdaq: CMPX.
