## TL;DR

MOMA Therapeutics makes tablets that block DNA repair machinery that some cancers depend on to survive. One medicine is for tumours with faulty BRCA-type repair, another for tumours with mismatch repair defects.

## Summary

MOMA Therapeutics, based in Cambridge, Massachusetts, targets molecular machines, enzymes that work through stepwise changes in shape, and launched in 2020, according to the company. MOMA-313 is an oral inhibitor of the helicase domain of DNA polymerase theta (Pol theta), in a phase 1 trial (NCT06545942) alone and with olaparib in homologous recombination deficient solid tumours. MOMA-341 is an oral inhibitor of the Werner (WRN) helicase, in a phase 1 trial (NCT06974110) alone or with irinotecan or immunotherapy in MSI-high or mismatch repair deficient tumours, with a first patient dosed in July 2025. In January 2025 the company in-licensed a selective PARP1 inhibitor, MOMA-989, for development alone and with MOMA-313. It announced a discovery collaboration with Roche in January 2024 and a collaboration and licence agreement with Bayer in October 2024. It raised a 150 million US dollar Series B in May 2022 led by Goldman Sachs Asset Management. Its first registered cancer trial, NCT06545942, was posted on ClinicalTrials.gov in August 2024.
