## TL;DR

Loki Therapeutics uses weakened Listeria bacteria to deliver a piece of the tetanus vaccine into tumour cells, so that immune cells primed by childhood tetanus vaccination recognise and attack the tumour. Its first trial, with gemcitabine, is in pancreatic cancer that cannot be removed by surgery.

## Summary

Loki Therapeutics is an immuno-oncology company whose AWAKE technology and Myeloid Mediated Delivery platform use an attenuated bacterial vector to carry childhood recall antigens, such as tetanus toxoid, selectively into tumours and metastases, so that existing memory T cells from tetanus vaccination target tumour cells displaying the antigen. Preclinical work in pancreatic cancer models with Listeria-delivered tetanus toxoid drew trade press coverage in March 2022. Its candidate Lm-LLO-TT is in a phase 1 study with gemcitabine in unresectable pancreatic ductal adenocarcinoma, run with the National Cancer Institute (NCT07828236), posted in September 2026 and not yet recruiting when read. The company's chief executive and co-founder is Christopher Bradley. Its first registered cancer trial, NCT07828236, was posted on ClinicalTrials.gov in September 2026.
